E85/E54?

Before I owned my 550i I had a 370z so excuse me if this is a stupid question. I want to run E85 or E54 in my 550i but I’m not sure if I would need anything upgraded or some sort of tune to run ethanol? I have the VRTune by Vivid Racing

Unless you've already exceeded the limitations of premium 91/93 octane knock resistance there is no reason to run ethanol blends.

JB4 Beta tune allows you to mix in E85.

Stock with no tune you can run up to 25% so E25

I think you're better off with 1/2 tank of Shell Vpower +nitro 93 and 2 cans of NOS booster

Ethanol blends are higher octane and definitely make a difference. But for E54 you are burning approx. 20% more fuel so you need to map and have a fuel system capable of delivering that. I am not sure on the 550i, but on the N55 our HPFP's can't deliver enough fuel so supplemental fueling is necessary. Ethanol blends also allow for more timing so you can get more HP at lower boost.

E20 is about the most you'll be able to go on the stock 550i which will be good for cooling/cleaning and timing corrections. I doubt you'd pick up any power. I run E20 in mine, but it runs much better with full 93 and 2 bottles of NOS Booster.
